Directed differentiation of human induced pluripotent stem cells to hepatic stellate cells

چکیده
Hepatic stellate cells (HSCs) are nonparenchymal liver responsible for extracellular matrix homeostasis and the main involved in development of fibrosis following injury. The lack reliable sources HSCs has hence limited complex vitro systems to model diseases toxicity. Here we describe a protocol differentiate human induced pluripotent stem (iPSCs) into hepatic (iPSC-HSCs). is based on addition several growth factors important sequentially over 12 d. iPSC-HSCs present phenotypic functional characteristics primary can be expanded or frozen used perform high-throughput studies. We also how coculture with hepatocytes, which self-assemble three-dimensional (3D) spheroids. This enables generation HSC-like modeling drug screening Human iPSCs differentiated by culture factors. They respond fibrogenic stimuli, arising as new source modeling. Subsequent hepatocytes facilitates self-assembly 3D

Induced pluripotent stem cells and hepatic differentiation.
Induced pluripotent stem cells (iPSCs) are generated by reprogramming somatic cells to a pluripotent state by the introduction of specific factors. They can be generated from cells of different origins, such as fibroblasts, keratinocytes, hepatocytes, and blood. iPSCs are similar to embryonic stem cells (ESCs) in several aspects, such as morphology, expression of pluripotency markers, and the a...
متن کاملDirected Differentiation of Human Pluripotent Stem Cells to Microglia
Microglia, the immune cells of the brain, are crucial to proper development and maintenance of the CNS, and their involvement in numerous neurological disorders is increasingly being recognized. To improve our understanding of human microglial biology, we devised a chemically defined protocol to generate human microglia from pluripotent stem cells. Myeloid progenitors expressing CD14/CX3CR1 wer...
متن کاملInduced Pluripotent Stem Cells: Challenges and Opportunities
Regenerative capacity of mammals is limited and can rarely regenerate a specific organ or tissue fully. Due to these limitations, regenerative medicine seeks efficient and safe cell sources for regeneration of damaged tissues and organs or treatment for incurable diseases. Human embryonic stem cells (HESCs) hold two important properties called self renewal and pluripotency. However, the use of ...
متن کاملDirected differentiation of induced pluripotent stem cells towards T lymphocytes.
Adoptive cell transfer (ACT) of antigen-specific CD8(+) cytotoxic T lymphocytes (CTLs) is a promising treatment for a variety of malignancies (1). CTLs can recognize malignant cells by interacting tumor antigens with the T cell receptors (TCR), and release cytotoxins as well as cytokines to kill malignant cells. It is known that less-differentiated and central-memory-like (termed highly re...
