Hepatic stellate cells (HSCs) are nonparenchymal liver responsible for extracellular matrix homeostasis and the main involved in development of fibrosis following injury. The lack reliable sources HSCs has hence limited complex vitro systems to model diseases toxicity. Here we describe a protocol differentiate human induced pluripotent stem (iPSCs) into hepatic (iPSC-HSCs). is based on addition several growth factors important sequentially over 12 d. iPSC-HSCs present phenotypic functional characteristics primary can be expanded or frozen used perform high-throughput studies.
